چکیده

The loss of enzyme activity during the process of purification is a major obstacle when unstable enzymes are used. Douzou and co-workers introduced the idea of using chromatography at subzero temperature in aqueous/organic solvent systems (Balny et al.. 1975). In the present paper we describe the apparatus and a method of using an aqueous/ organic solvent system to purify unstable enzymes at subzero temperatures. The nuclear enzyme poly(ADP4bose) polymerase from pig thymus was used, which catalyses the sequential transfer of the ADP-ribose moiety of NAD+ to nuclear proteins [for reviews see Hilz &Stone (1976) and Hayaishi & Ueda (1977)l. I t is a very unstable enzyme, with a half-life of 15-20h at 4°C (Brightwell et al., 1975). A comparatively simple apparatus has been used that can achieve temperatures down to -25°C.

Formation and stability of Vitamin E nano-emulsion based delivery systems by spontaneous emulsification method: Glycerol as a co-solvent

There is considerable interest in using nano-emulsions (NEs) as delivery systems for lipophilicbioactive ingredients. In this study, the influence of G on the properties of vitamin E (VE) enriched NEsprepared using spontaneous emulsification (SE) method, were investigated. The particle size and optical clarityof the NEs depended strongly on the co-solvent presence, absence, an...

Energy Study at Different Temperatures for Active Site of Azurin in Water, Ethanol, Methanol and Gas Phase by Monte Carlo Simulations

The interaction between the solute and the solsent molecules play a crucial role in understanding the various molecular processes involved in chemistry and biochemistry, so in this work the potential energy of active site of azurin have been calculated in solvent by the Monte Carlo simulation. In this paper we present quantitative results of Monte Carlo calculations of potential energies of ...

Improving the Stability of Cold-Adapted Enzymes by Immobilization

Cold-adapted enzymes have gained considerable attention as biocatalysts that show high catalytic activity at low temperatures. However, the use of cold-adapted enzymes at ambient temperatures has been hindered by their low thermal stabilities caused by their inherent structural flexibilities. Accordingly, protein engineering and immobilization have been employed to improve the thermal stability...

Non-Aqueous Biocatalysis in Homogeneous Solvent Systems

Enzymes are highly specific catalysts that typically function in aqueous solvents. However, many enzymes retain their catalytic activities at high concentrations in non-aqueous environments, including neat hydrophilic organic solvents. In fact, enzymes can be used to carry out reactions in organic solvents that are not possible in aqueous systems. Therefore, biocatalysis in homogenous non-aqueo...

Increasing Performance and Thermostability of D-Phenylglycine Aminotransferase in Miscible Organic Solvents

Background: D-Phenylglycine aminotransferase (D-PhgAT) is highly beneficial in pharmaceutical biotechnology. Like many other enzymes, D-PhgAT suffers from low stability under harsh processing conditions, poor solubility of substrate, products and occasional microbial contamination. Incorporation of miscible organic solvents into the enzyme’s reaction is considered as a solution...
